Background: The manufacturing of novel active pharmaceutical ingredients (APIs) often leads to materials the solubility and bioavailability of which is poor and, when treated by traditional formulation approaches, their in vivo performance is found to be problematic. Hot-melt extrusion (HME) has recently attracted a lot of attention in the pharmaceutical industry, as it can be employed to engineer such materials, by forming amorphous solid dispersions, and tackle the aforementioned issues. The use of HME also bears the advantage of being suitably coupled to 3D printing equipment which can in turn be used to produce personalised medicine.

Scope: This project will first focus on reviewing various HME mechanistic modelling approaches and the most suitable ones will be implemented in MATLAB. The model(s) will then be employed to perform sensitivity analysis and parametric studies in order to investigate the effect of the model inputs and design parameters on the system performance.
